Did you know that estrogen can be metabolized by several pathways into different metabolites?
- 2-Hydroxyestrone metabolite – weaker estrogen receptor binding affinity, protective
- 16α-Hydroxyestrone metabolite – strong estrogenic activity, stimulates estrogen receptor
- 4-Hydroxyestrone metabolite – potent, can oxidize to quinones; potentially pro-carcinogenic
Estrogen metabolized to 4-OHE1(E2) can lead to the formation of DNA adducts. This process is a putative tumor initiating event.
